prepacked DEAE anion exchange column?

Hi,

I would appreciate any suggestion about where I can buy a reliable pre-packed DEAE anion exchange column for protein and DNA analysis. I need to use that in Perkin Elmer autosampler HPLC system so I have to be cautious about packing it by myself since I have not done that before.

PolyLC sells PolyWAX LP columns. While the functional group is not DEAE-, this material is still a weak anion-exchange (WAX) material, which is the relevant concern for your two applications. You should get a material with a pore diameter of either 1000- or 1500-Å (the 1500-Å material is particularly effective for PCR reaction product purification).

A DEAE- functional group is a tertiary amine; it stands for "diethylaminoethyl-". A material with DEAE- groups is a weak anion-exchange material (WAX-type). Strong anion-exchange (SAX) materials have quaternary amine functional groups. The difference in performance (other than the more hydrophobic character of a SAX material) is that WAX materials lose their (+) charge in a continuum from pH 5 to pH 9.5, while SAX materials retain most of their charge as high as pH 10. Now, which kind of material does Sepax make?
