I'm trying to set up a method for the identification of anthocyan on grape juice, i'm unsing and C18 for the separation (250x2.0) and a LCQ for the identification.The mobile phase is a gradient of water and ACN both with 1% of formic acid.
I'm having a little problem on the TIC of the LCQ, the chromatogram looks quite good but the ion accumulation time does not seem to change across the width of a peak. I veified the the spray and look ok even if my spray current is quite high ranging around 25 at the departur of the begginig with a lot of aqueous.
i'm wondering if my flow is too high for the LCQ, i'm working at 0.4ml/min, what would be the ideal flow for the LCQ?
